If you mean a remote filesystem mounted with NFS, yes, there will be 
problems. You could use a NAS box with courier-imap, since courier 
doesn't need to lock files, but with Cyrus that will not work well at 
all. You'll get better performance with Cyrus since it keeps indexes of 
practically everything, but at the cost of not being able to use an 
NFS-mounted filesystem.
